Rune tiles are crafted using alchemy, and can be put together into tilesets to gain damage and healing buffs in combat. Most of the individual runes are based on the Elder Futhark alphabet.

Attack Speed Runes
These tiles are triggered by hitting an enemy multiple times in the span of a second.
DoT ticks like Poison, Affliction or Bleed do not count as hits.
The game keeps track of the hits per second with a background clock - this means that, for example, an attack speed of 2.2 hits per second can trigger either ᛗ or ᚾ, depending on when the "second" starts.
If more than one enemy is hit within a second, the Attack Speed runes won't activate, Attack Multiple Targets Runes will be triggered instead.
Blocking an attack with the shield counts as a hit on the attacking enemy. Getting attacked by multiple enemies will trigger Attack Multiple Targets Runes.
Musketeer:
Normal shots count as two hits.
The turret healing/damaging counts as a hit.
A shot with Poison, Weakness, Gravity and Impact counts as 3 hits.
Charged shots with the Charged Orb talent count as hits on all enemies they reach (even if no orb is used) and will trigger Attack Multiple Targets Runes instead.
Ranger:
Charged Shots counts as two hits.
Poison Arrows count as two hits (three if charged).
Mage:
Arcane Explosion counts as two hits if a single target is hit. If more than one enemy is hit, Attack Multiple Targets Runes will trigger instead.

Attack delay runes trigger immediately before the next hit. For example, starting combat by using a Piercing arrow will trigger ᚦᚨ.
Delay runes work off of actual time since last hit, and do not depend on the background clock like the Hits Per Second runes.
